About Urolithin A
Urolithin A is a bioactive compound derived from foods like pomegranates. In skin health, it primarily functions by activating cellular repair pathways, notably supporting proteins like SIRT1 that help protect and maintain DNA integrity. Health Benefits
Skin cell biological clocks are crucial for regulating repair, regeneration, and barrier function. First identified in 2000 by American biologist Susanna B., the clock genes CLOCK and PER1 were shown to drive daily physiological cycles. Disruption of this rhythm increases water loss, impairs barrier function, and accelerates collagen breakdown.
Studies show that Urolithin A can restore and strengthen circadian rhythms: it boosts the amplitude of luciferase oscillations in aged cells by up to fourfold, normalizes clock gene expression such as BMAL1 and PER2 in intestinal epithelial cells, and modulates the central clock system in the suprachiasmatic nucleus (SCN) of mice.

Studies show that Urolithin A (UA) treatment significantly enhances collagen deposition and remodeling in wounds. Masson’s trichrome staining and collagen I/III immunohistochemistry showed stronger, regularly arranged collagen fibers in UA-treated groups compared to controls.
Moreover, UA accelerates re-epithelialization, as indicated by thicker neonatal epidermis and increased cytokeratin expression, suggesting faster wound closure and improved skin regeneration.

In a full-thickness skin wound model, Urolithin A (UA)-treated wounds healed faster than controls. By day 7, the UA group achieved nearly 70% wound closure compared to 50% in the control group, and by day 21, wounds in the UA group were almost fully closed.
Mechanistic studies revealed that UA binds directly to PI3K and AKT, inhibiting their phosphorylation and regulating the PI3K/AKT signaling pathway. This promotes angiogenesis and collagen deposition, supporting faster wound healing and enhanced skin cell renewal.
